Magnesium (as Magnesium Bisglycinate Chelate – TRAACS™)

Magnesium is essential for more than 300 enzymatic reactions related to protein synthesis, glucose balance, muscle and nerve function, and healthy blood pressure. Magnesium bisglycinate is highly bioavailable and has been studied for its ability to reduce dysmenorrhea by relaxing uterine smooth muscle and lowering prostaglandins that cause cramping. Magnesium levels often drop during the second half of the menstrual cycle, and supplementation helps maintain optimal balance. It also supports thyroid function—both in forming T4 (thyroxine) and converting T4 to the active hormone T3, which drives metabolic activity. Low magnesium has been associated with increased thyroid antibodies and higher risk of Hashimoto’s and hypothyroidism.

Iodine

Iodine is naturally present in foods like kelp and is crucial for producing T3 and T4 thyroid hormones, as well as TSH from the pituitary. These hormones regulate metabolic rate, protein synthesis, and numerous biochemical pathways. About 70% of the body’s iodine is found in hormone-sensitive tissues such as the ovaries, uterus, and breasts. Adequate iodine supports ovulation, reduces estrogen dominance symptoms (heavy periods, breast tenderness, irritability), may reduce ovarian cyst formation, and helps the body detoxify estrogen metabolites.

L-Tyrosine

Tyrosine is a conditionally essential amino acid required for the synthesis of dopamine, norepinephrine, and epinephrine, as well as T3 and T4. Thyroid hormones are literally tyrosine molecules bound to iodine, making tyrosine foundational for thyroid function. Supplementation may help support thyroid hormone levels, although individuals with overactive thyroid or those on thyroid medication should consult a clinician before use.

Turmeric Extract (95% Curcuminoids)

Curcumin, the active compound in turmeric, has been extensively studied for its powerful anti-inflammatory and antioxidant effects. It inhibits inflammatory enzymes and the NF-κB pathway—central to cytokine production, tissue destruction, and autoimmune responses. By reducing inflammation, curcumin may help support individuals with autoimmune thyroid conditions such as Hashimoto’s. It can also positively influence PMS symptoms by improving neurotransmitter levels (dopamine, norepinephrine, serotonin), which naturally decline with falling estrogen and progesterone. Because curcumin alone has low bioavailability, POISE includes Bioperine®, which has been shown to increase curcumin absorption by up to 2000%.

Olive Leaf Extract (20% Oleuropein)

Olive polyphenols, especially oleuropein, are potent antioxidants with demonstrated anti-inflammatory and immune-supporting benefits. These properties extend to thyroid and PMS support. Oleuropein helps stabilize free radicals, reduce oxidative stress, and limit lipid oxidation. Research also suggests olive leaf extract may improve thyroid function by lowering elevated TSH and increasing T3 levels in individuals with underactive thyroid.

Chaste Tree Extract (Vitex agnus-castus – 1.5% Agnusides)

Chaste berry has a long history of use for women’s hormonal health. Clinical reviews support its efficacy for PMS, menstrual irregularities, dysmenorrhea, luteal phase insufficiency, hyperprolactinemia, menopausal symptoms, acne, and more. Vitex increases dopamine activity, which reduces prolactin levels. High prolactin can disrupt estrogen and progesterone balance and cause menstrual irregularities. Clinical trials show that Vitex can reduce PMS symptoms when taken consistently, though symptoms may return when the herb is discontinued.

Setria® L-Glutathione

Glutathione is the body’s master antioxidant, synthesized from glutamic acid, cysteine, and glycine. During PMS, higher stress and fluctuating hormone levels can increase free radical production. Setria® glutathione helps neutralize these radicals and supports the body’s antioxidant systems. It also recycles vitamins C and E back into their active antioxidant forms after they neutralize free radicals, amplifying overall cellular protection.

3,3'-Diindolylmethane (DIM)

Estrogen exists in multiple forms (E1, E2, E3) and is metabolized into compounds that can have either beneficial or harmful effects. DIM supports a healthier estrogen profile by: Increasing production of protective estrogen metabolites (like 2-OHE) Reducing production of more potent, proliferative metabolites (like 16-OHE1) Modulating cytochrome enzymes to reduce estrogen receptor overstimulation Reducing unwanted cell proliferation associated with certain estrogen pathways DIM is a stable, highly effective metabolite of indole-3-carbinol (I3C) and delivers all of its hormonal-balancing benefits without the instability of I3C itself.

Pregnenolone

Known as the “grandmother hormone,” pregnenolone is the precursor to progesterone, estrogen, and many other steroid hormones. Low pregnenolone—often due to chronic stress—can reduce progesterone production, contributing to estrogen dominance, heavy periods, low libido, and weight gain. By supporting progesterone balance, pregnenolone may help reduce PMS severity and promote more stable hormonal rhythms throughout the menstrual cycle.

BioPerine® (Black Pepper Extract – 95%)

BioPerine® enhances nutrient absorption through improved intestinal blood flow and slowed intestinal motility. In Core POISE, its primary purpose is to dramatically increase the bioavailability of curcumin, with studies showing more than a 500% improvement in serum levels. It has also been shown to increase selenium absorption significantly, further supporting overall metabolic and antioxidant function.
